Description
The Water DNA Isolation kit provides a quick and easy method for the extraction of genomic DNA from water samples from various sources. A large number of microorganisms exist in water samples, and these microorganisms are widely used as important ecological indicators. The extraction of high-purity genomic DNA from water is a very important method for the detection of water environmental values.
Water samples contain a large number of inhibitory factors such as humic acid, metal ions, etc. Even if these substances exist in trace amounts in the purified DNA, they will affect downstream reactions, such as PCR, restriction enzyme digestion, etc. Therefore, the key to purifying genomic DNA in water is how to effectively remove the inhibitory factors in water. Using the company's unique DNA-only silica membrane spin column and solution formulation, with Foregene Protease, it can effectively remove various inhibitory factors in water without organic solvent extraction or ethanol and isopropanol precipitation, within 40 minutes. Complete the extraction of DNA from water samples.

Kit components
- Lysozyme: enzymatically hydrolyze the cell wall of positive bacteria.

Genomic DNA fragment size
The water body genomic DNA extraction kit uses silica gel membrane column to separate and purify the water body genomic DNA, and the size of the purified genomic DNA fragments are all around 23kb. As shown in the figure:
Notes: (Please read the notes carefully before using the kit)
- The single genomic DNA extraction of each water body sample should be determined according to the microbial content and turbidity in the water body, and the maximum amount should not exceed 30L.
-Before use, carefully check whether there is precipitation in Buffer SG2, Buffer SG3, Buffer SG4 and Buffer PW. If there is precipitation, please dissolve it at 37°C, mix well before use.
- Before using the kit, be sure to check that BufferWB has been added with absolute ethanol as instructed. Add 6ml absolute ethanol (DE-05611), 30ml absolute ethanol (DE-05612), 60ml absolute ethanol (DE-05613), 120ml absolute ethanol (DE-05614), 300ml absolute ethanol to BufferWB before use (DE-05615).
- During the sample lysis process, always keep the sample immersed in the lysis buffer. If the sample sticks to the cap and inner wall of the tube, it can be processed by brief centrifugation.
- Elution volume: Buffer EB should not be less than 50μl, otherwise it will affect the DNA yield.
- Remember not to add RNase to any buffer.
- All centrifugation steps are performed using a tabletop centrifuge at room temperature (15-25°C).
- All experimental steps were carried out at room temperature (15-25°C).
Storage and Shelf life
-The water DNA Isolation kit can be stored for 12 months under dry conditions at room temperature (15-25°C), if it needs to be stored for a longer period of time, it can be stored at 2-8°C.
Note: If stored at low temperature, the solution is prone to precipitation. Before use, be sure to put the solution in the kit at room temperature for a period of time. If necessary, preheat it in a 37°C water bath for 10 minutes to dissolve the precipitate, and mix it before use.
-Foregene Protease solution has a unique formula, which is active when stored at room temperature for a long time (3 months); when stored at 4℃, its activity and stability will be better, so it is recommended to store it at 4℃, remember not to store it at -20℃.
-Dry powder Lysozyme is stored at -20°C; the prepared Lysozyme solution is divided into small portions and stored at -20°C.
