Plant Seed Direct PCR Kit adopts a unique cleavage reaction system, which can rapidly release genomic DNA from plant seed samples with low polysaccharide and polyphenol content (such as: rice, wheat, tobacco, corn, soybean, etc.) for PCR reaction, so it is especially suitable for Large-scale genetic testing. In order to meet the needs of different detection experiments, this kit can use various types of samples (such as whole seeds, micro-tissue cut or ground samples of multiple seeds) for experiments. Among them, for seed samples that still need to be germinated, micro-tissue pieces (1-5 mg) other than seed embryos can be selected for experiments; for experiments that require sampling and testing in a large number of samples, multiple samples can be mixed and combined. After grinding into particles with a diameter of about 0.5mm, the experiment is carried out (at least 0.1% of the target sample can be detected).

The cleavage reaction system can release genomic DNA within 5-10 minutes, without the need for other processes of deproteinization, RNA or secondary metabolites, and the released trace DNA can be used as a template for PCR reaction.

2× Seed PCR Easy^TM Mix has a strong resistance to PCR reaction inhibitors, and can directly use the lysis mixture of plant material as a template for efficient and specific amplification. This reagent contains the company's D-Taq DNA Polymerase, dNTPs, MgCl2, reaction buffer, PCR reaction enhancer, optimizer and stabilizer specially designed for direct PCR modification. It can be used in combination with the cleavage reaction system to detect samples quickly and easily, and has the characteristics of high sensitivity, strong specificity and good stability.

D-Taq DNA polymerase is a DNA polymerase specially developed by Foregene for direct PCR reactions. D-Taq DNA polymerase has strong tolerance to a variety of PCR reaction inhibitors, and can efficiently amplify trace amounts of DNA in various complex reaction systems. The amplification speed can reach 2Kb/min. It is especially suitable for Direct PCR reaction.

According to the differences of plant samples, the products are divided into two categories: Plant Seed Direct PCR Kit and Plant Seed Direct PCR Plus Kit. In the lysis stage, considering that different types of seeds or tissue materials have large differences in the amount of reagents used in the lysis process, customers can choose the corresponding plant seeds (small and medium-sized) direct PCR kit or plant seeds (large) according to the size of their experimental materials. Direct PCR kit; in the PCR detection stage, customers can also choose the common 2× Seed PCR Easy^TM Mix or the 2× Seed PCR Easy^TM Mix (UNG) which has the effect of preventing the contamination of PCR amplification products according to the experimental needs.

Kit Component Information

- Buffer SP1: Provides the environment required for plant seed lysis reactions.

- Buffer SP2: Neutralizes the lysate so that it does not interfere with subsequent PCR reactions.

- 2× Seed PCR EasyTM Mix: Contains Taq DNA Polymerase specially modified by Fuji Bio, dNTPs, MgCl2, reaction buffer, PCR reaction enhancer, optimizer and stabilizer, etc. For PCR reactions, simply add the appropriate lysis mix, primers, and ddH2O to the 2× Seed PCR Easy^TM Mix for PCR reactions.

- 6× DNA Loading Buffer: This Loading Buffer does not contain SDS. It is recommended to use the 6× DNA Loading Buffer provided with the kit when performing agarose gel electrophoresis in order to obtain good electrophoresis results. Do not use Loading Buffer containing SDS, otherwise there will be a large tail of light in the swimming lane during electrophoresis, which will affect the experimental results.

Storage and Shelf life:

-Part I of this kit should be stored at room temperature or 2-8℃.

vReagents Buffer SP1, Buffer SP2, 6× DNA Loading Buffer, under dry conditions, can be stored for 12 months. If you need to store the kit for a longer time, you can store it at 2-8℃.

-Part II of this kit should be stored at -20℃.

vReagent 2× Seed PCR Easy Mix, if used frequently, it can also be stored at 4℃ for short-term storage (use up within 10 days).
