Description
This product uses a unique lysis reaction system, which can quickly release genomic DNA from plant seed samples with low polysaccharide and polyphenol content (such as: rice, wheat, tobacco, corn, soybeans, etc.) for use in PCR reactions, so it is particularly suitable Large-scale genetic testing. In order to meet the needs of different detection tests, this kit can use multiple types of samples (such as whole seeds, small tissue cuts, or ground samples of multiple seeds) for experiments. Among them, for seed samples that still need to be germinated, you can choose to cut out micro-tissue cuts (1-5mg) other than the seed embryo for experiment; for experiments that need to be sampled in a large number of samples, you can choose to mix multiple samples and combine them. After grinding into particles with a diameter of about 0.5mm, the experiment can be carried out (at least 0.1% of the target sample can be detected).
The process of releasing genomic DNA from the cleavage reaction system can be completed within 5-10 minutes. There is no need for other processes to remove protein, RNA or secondary metabolites, and the released trace amounts of DNA can be used as a template for PCR reactions.
2× Seed PCR EasyTM Mix has strong tolerance to PCR reaction inhibitors, and can directly use the lysis mixture of plant materials as a template for efficient and specific amplification. This reagent contains the company's D-Taq DNA Polymerase, dNTPs, MgCl2, reaction buffer, PCR reaction enhancer, optimizer, and stabilizer specially modified for direct PCR. Used in conjunction with the lysis reaction system can quickly and easily detect samples, and has the characteristics of high sensitivity, strong specificity, and good stability.
2× Seed PCR EasyTM Mix (UNG) uses dUTP instead of dTTP on the basis of 2× Seed PCR EasyTM Mix, and adds UNG enzyme (Uracil-N-glycosylase) that can degrade the template containing dUTP at the same time. Before the PCR reaction, the UNG enzyme is used to degrade the PCR product containing uracil. The UNG enzyme will not have any effect on the template that does not contain uracil, thereby ensuring the specificity and accuracy of amplification and preventing the possibility of large-scale genetic testing. Contamination of PCR products occurred D-Taq DNA polymerase is a DNA polymerase specially developed by Foregene for direct PCR reactions. D-Taq DNA polymerase has strong tolerance to a variety of PCR reaction inhibitors, and can efficiently amplify trace amounts of DNA in various complex reaction systems, and the amplification speed can reach 2Kb/min, which is especially suitable for Direct PCR reaction.

Storage

Note: If stored at low temperature, the Buffer SP1 solution is prone to precipitation. Before use, be sure to place the solution in the kit at room temperature for a period of time. If necessary, preheat it in a 37°C water bath for 10 minutes, wait for the precipitation to disappear, and shake the solution before use.
Part II should be stored at -20°C.
