Fast room temperature operation Stool DNA Isolation kits Extraction kits for genomic dna extraction
Description
This kit provides a quick and easy method to extract genomic DNA from bacteria and food residues from stool samples from various sources. There are a large number of inhibitors in stool samples. Even if these substances are present in a small amount in the purified DNA, they will affect downstream reactions, such as PCR and restriction enzyme digestion. Therefore, the key to purification of fecal DNA is how to effectively remove inhibitors in feces.
This kit uses DNA-Only Column that can specifically bind DNA, a brand-new Foregene Protease and a unique buffer system, which can effectively remove various inhibitors in feces, without organic solvent extraction or ethanol and isopropanol precipitation. The DNA extraction operation from stool samples can be completed in 40 minutes.

Features&advantages
- No RNase contamination: The DNA-Only Column provided by the kit makes it possible to remove the RNA from the genomic DNA without adding RNase during the experiment, preventing the laboratory from being contaminated by exogenous RNase.
- Fast speed: The operation is simple, and the fecal genomic DNA extraction operation can be completed within 40 minutes.
- Convenient: The centrifugation is performed at room temperature, and there is no need for 4°C low-temperature centrifugation or ethanol precipitation of DNA.
- Safety: No organic reagent extraction is required.
- High quality: The extracted genomic DNA has large fragments, no RNA, no RNase, and extremely low ion content, which can meet the requirements of various experiments.

◆ Buffer SL2: to remove grease impurities.
◆ Buffer SL3: Inactivate protease and provide a DNA loading environment.
◆ Buffer PW: Remove impurities such as protein and RNA in DNA.
◆ Buffer WB: Remove residual salt ions in DNA.
◆ Buffer EB: elute the DNA on the purification column membrane.
◆ DNA-only Column: Specific adsorption of genomic DNA in the lysate.
DNA-Only Column Characters
Maximum binding capacity | 80μg |
Maximum loading volume | 800μl |
Longset DNA fragment | 23kb |
Minimum elution volume* | 60μl |
Selection of samples | Newly taken stool samples |
Maximum amount of starting material* | 200mg stool sample |
*: The minimum elution system of 60μl is a reasonable recommended volume considering DNA recovery and concentration. If you want to increase the yield of DNA, you can appropriately increase the volume of the eluate; if you want to increase the concentration of purified DNA, at the expense of part of the DNA yield, reduce the volume of the eluate appropriately, such as using a 40μl elution system, In order to get a higher concentration of DNA.
Genomic DNA extraction yield and purity
The amount of fecal genomic DNA extracted is related to factors such as the source, storage time, and moisture content of the fecal sample. Stool DNA Isolation Kit is used to process stool samples from various sources, and the amount and purity of genomic DNA obtained are shown in the following table:
Sample source | Sample dosage(mg) | DNA yield(μg) | OD260/280 | OD260/230 |
cattle | 200 | 4-6 | 1.7-1.9 | 1.7-1.9 |
Domestic pig | 200 | 4-6 | 1.7-1.9 | 1.8-1.9 |
Poultry (chicken) | 200 | 4-6 | 1.7-1.9 | 1.7-1.8 |
Note: The data in this table is for reference only. In actual operation, due to the storage conditions of the materials used, operating proficiency and other factors, the data obtained may differ slightly from the data in this table.
Genomic DNA fragment size
Stool DNA Isolation Kit uses silica gel membrane columns to separate and purify fecal genomic DNA. If the sample is stored well or the sample is fresh, the purified genomic DNA fragment size is around 23kb; if the sample is stored for a long time or the operation is improper during the extraction process, It will cause the genomic DNA to degrade into diffuse bands. As shown in the figure:
Storage and Shelf life
- This kit can be stored for 12 months under dry conditions at room temperature (15-25°C); if it needs to be stored for a longer period of time, it can be stored at 2-8°C.
Note: If stored at low temperature, the solution is prone to precipitation. Before use, be sure to place the solution in the kit at room temperature for a period of time. If necessary, preheat it in a 37°C water bath for 10 minutes to dissolve the precipitate, and mix it before use.
- Foregene Protease solution has a unique formula, which is active when stored at room temperature for a long time (3 months), its activity and stability will be better when stored at 4°C, so it is recommended to store it at 4°C, remember not to keep it at -20°C.
- Dry Lysozyme powder is stored at -20°C, the prepared Lysozyme solution is divided into small portions and stored at -20°C.
